- Due to the accumulation of crystals on the surface of hematoxylin in the staining solution, filter it through filter paper before use. Handle with care when dyeing.
- Before use, it should be thoroughly mixed, and during the dyeing process, the numerals of the dyeing frame should be lifted and rotated. Update after 500-800 slides per cylinder.
The effect of hydrochloric acid differentiation is to differentiate the hematoxylin on the cytoplasm, so the time should not be too long. But if differentiation is not possible, it will result in the nucleus being too dark in color or (and) the cytoplasm being difficult to color. Immediately rinse with tap water after differentiation. Not suitable for prolonged storage, otherwise all colors may disappear.
4. Alkalization, also known as the process of returning to blue, results in a shorter return to blue and insufficient composition of hematoxylin, which then affects the staining effect. Each cylinder of lithium carbonate is updated with 500 glass slides; The frequency of changing warm water should be higher.
5. The orange dyeing time should not be too long, otherwise it is not easy to color. Orange color updates with 500 glass slides per vat.
